چکیده

Minute intestinal flukes, Haplorchis taichui and H. pumilio, belong to the family Heterophyidae (Trematoda: Platyhelminthes), which have been studied very limited, especially molecular markers of mitochondrial genomes (mtDNA) ribosome transcription units (rTU or rDNA). We obtained complete genome coding part unit pumilio Vietnam. Nucleotide amino acid data were compared between Metagonimus yokogawai for genomic/gene composition, codon usage (skew/skewness), tandem repeat (TRU). The mtDNA (strain Htai-QT3-VN) with length 15,120 bp M. (15,258 bp; Korea; KC330755) contains 36 genes, including 12 protein-coding genes (cox1, cox2, cox3, nad1, nad2, nad3, nad4L, nad4, nad5, nad6, atp6 cob), 2 ribosomal RNA (rRNA); 22 transfer (tRNA trn) a noncoding region (NCR) trnE trnG, divided into sub-regions containing 5 TRUs (182–183 bp/TRU). (Vietnam Laos) uses A = 19.56%, T 39.71%, G 28.34%, C 12.39% (A + is 59.27% and 40.73%) construction, whose skew/skewness value at A+T negative (–0,340) G+C positive (0.392); (PCGs) similar; but mito-ribosomal (MRGs, 16S/rrnL 12S/rrnS) it less (57.22%) more (42.78%). had lower (mtDNA/55.68%; PCGs/55.96%; MRGs/54.15%) higher rate than taichui. Vietnam Laos has 10,164 encoding 3,376 acids construct PCGs mostly used codons as Phenylalanine (Phe-TTT) Leucine (Leu-TTG), leastly codonsas Glutamine (Gln-CAA), Arginine (Arg-CGC). Additional condon, Thr-ACA/ACC can be added least in yokogawai. rTU (from '18S 3' 28S) (7,268 bp) (7,416 identified genomic regions 18S rDNA, ITS1, 5.8S ITS2 28S rDNA. both species same (1,992 18S, 160 5.8S), different (3,875 3,870 pumilio). ITS1 (797 (280 do not contain TRUs, whilst (1,106 TRUs(136 3 TRU 116 TRUs); (444 (83–85 bp/each).

Complete Mitochondrial Genome of Haplorchis taichui and Comparative Analysis with Other Trematodes

Mitochondrial genomes have been extensively studied for phylogenetic purposes and to investigate intra- and interspecific genetic variations. In recent years, numerous groups have undertaken sequencing of platyhelminth mitochondrial genomes. Haplorchis taichui (family Heterophyidae) is a trematode that infects humans and animals mainly in Asia, including the Mekong River basin. We sequenced and...

In vitro development of Haplorchis taichui (Trematoda: Heterophyidae).

Newly excysted metacercariae of Haplorchis taichui were cultured in a candle jar set at 37 degrees C. Both monophasic culture media [0.85% NaCl, RPMI 1640, RPMI 1640+10% fetal calf serum (FCS)] and diphasic culture media [RPMI 1640 + egg yolk agar, RPMI 1640 + 5%, 10% or 15% blood in blood agar (BA), RMPI 1640 + 5%, 10% and 15% FCS with 5% blood in BA] were used in vitro. Parasites survived for...

In vitro excystation of Haplorchis taichui (Trematoda: Heterophyidae).

The effects of trypsin, bile extract, temperature and acid-based condition for the in vitro excystation of Haplorchis taichui metacercariae were studied. At 37 degrees C, approximately half the number of metacercariae excysted when exposed to 1% trypsin for 15 minutes with no more excystation found beyond this time. Increasing trypsin concentration seemed to reduce the excystation rate while bi...

Recovery and growth of Haplorchis taichui (Trematoda: Heterophyidae) in chicks.

An experimental study was performed to observe the recovery and growth of a minute intestinal fluke, Haplorchis taichui in chicks (Gallus gallus domesticus). Metacercariae of H. taichui were isolated from Jullien's mud carp, Henicorhynchus siamensis, which were collected in the Chiang Mai Province, Thailand. Two hundred metacercariae were orally force-fed to each chick. The intestine of the chi...

Surface ultrastructure of excysted metacercariae of Haplorchis taichui (Trematoda: Heterophyidae).

The morphology of newly excysted juvenile Haplorchis taichui was studied using a light and a scanning electron microscope (SEM). The reproductive organs were well developed. The whole body surface was covered with numerous transverse rows of scale-like spines, which had 3-11 points at the tip. The spines on the dorsal were similar with those on the ventral surfaces in shape, size and number of ...
